It started as a small enterprise running between The Hub and Junktown. In 2260, Duke met Albert at a bar in Junktown, and hired him as a bodyguard, with no money to pay him; he gave Albert 25% of the entire Company. Royal Duke Limited slowly grew and became more successful, until Royal Duke Limited was travelling to the Boneyard and Maxson via Necropolis . During a period in 2263 when raiding and banditry was an all time high, Hunter made his services available to Duke, who took him on immediately. He met Growler in 2267, and took him on as a sort of apprentice. Duke moved his company to the Mojave in 2270. Almost as soon as he and the boys arrived in the Mojave, they were attacked by a Legion Raiding Party. This led to Crow pledging himself to Duke. In 2280, Sandy and her gang attacked the caravan, and she was also hired. To this day, Royal Duke Limited is a trading company that travels across the Mojave, of course seriously belittled by the might of Crimson Caravan, Duke and his crew are still popular around the settlements across the great desert.

Alrighty a few things... The biggest of which would be their size in comparison to their reputation. You said they were a major trading caravan... But they have barely enough people for two caravans. That's barely enough to set up a regular trading route between nearby settlements, let alone the distance between The Hub and Vault City which is, I'm guessing, about 300 miles. Given the human capacity to walk 30-80 miles in a day, given training and what not... they'd make the trip to and from the place in three weeks, provided there is no interference... So you don't have nearly enough people for that kind of trip... Not unless they were just free lance travelling merchants.
You do realise that whilst in NCR territory, they'd have to use NCR dollars right? I imagine most early NCR settlements probably use nothing but NCR dollars. So just keep that in mind.
As for the enemies, I imagine the Crimson Caravan would be their competitive enemy and as we know with Cass, they'll probably try their best to put your guys out of business.

They use NCR dollars, they just don't like them very much. They rip easily, and if your not careful the brahmin eat it

This is approvable and moveable. But beware: Dukes is not without its competition. Alison McLafferty has the backing of the NCR, and Cass has lots of whiskey and a gun.
